Oh, wow... Hard one. I would say that knowing the person well, one would definitely sense the difference. Otherwise, just by intonation and perhaps a slight smirk on one's face ))

It depends on the question itself. Optimism is more linked with emotions whereas cynicism is more in line with "self-interest". To answer your question, I might say it comes down to the question and if it is asked through emosion or pure thought.
Optimism and cynicism are two different attitudes towards life and the world.
